An analysis of cerebrospinal fluid (CSF) from the phase 3 VALOR trial identified 56 proteins that changed in participants treated with tofersen compared with placebo. GPNMB increased consistently after treatment, with changes detectable as early as four weeks. The finding was also confirmed in separate tofersen-treated groups.
Why this matters
GPNMB could help researchers measure how people with SOD1-related amyotrophic lateral sclerosis (SOD1-ALS) respond biologically to tofersen. This is a pharmacodynamic finding, not evidence that GPNMB itself improves health or that treatment decisions should change.
