In a Drosophila model, glial cells producing C9orf72-related repeat products showed toxicity and reduced lifespan. The GR dipeptide repeat and the full G4C2 repeat caused the strongest effects and activated an endogenous retrovirus. Removing affected glial cells partly improved lifespan effects, while blocking cell death worsened them.
Why this matters
The findings suggest that glial cells may contribute to C9orf72-related ALS and frontotemporal dementia mechanisms. This is an animal and mechanistic study, so it does not establish the same effects in people or change current treatment.
Limitations and context
The work used fruit flies with engineered, cell-specific expression rather than people with ALS or frontotemporal dementia. It tested disease mechanisms and lifespan, not a treatment. The study also found that glial toxicity did not cause loss of nearby neurons in this model, and its mechanisms may differ from those in TDP-43 models.
