Researchers identified four compounds that activate valosin-containing protein (VCP) and increased clearance of insoluble TDP-43 aggregates in laboratory models of multisystem proteinopathy. The work links reduced nuclear VCP function with impaired protein clearance in cells carrying disease-associated VCP variants.
Why this matters
The findings may help explain how VCP variants contribute to multisystem proteinopathy, which can include muscle, bone and frontotemporal dementia features. This is early laboratory research and does not show that the compounds treat amyotrophic lateral sclerosis (ALS), multisystem proteinopathy or any other disease in people.
Limitations and context
The study was conducted in engineered cell lines and other multisystem proteinopathy models, not in people. It tested whether compounds affected protein aggregates and did not establish clinical benefit, safety or an effective dose. Further research would be needed to determine whether VCP activation helps patients.
